Course details

Covers the use of LC-MS in chemical analysis. The course starts with a general discussion of the theory of mass spectrometry as applied to the field of LC-MS. Types of LC-MS including LC-MSn are then compared. Subsequent sections of the course cover topics that relate to more advanced aspects of LC-MS including instrument components, modes of operation and the interpretation of LC-MS spectra. Other topics discussed include the optimization of quantitative and qualitative analyses using LC-MS and routine troubleshooting and maintenance of selected LC-MS instruments. Laboratory exercises provide practical training of the principles covered in the lectures.

Learning Outcomes

Upon successful completion, the student will be able to:

  • Demonstrate a thorough knowledge of the general theory of mass spectrometry where relevant to the field of liquid chromatography / mass spectrometry (LC/MS).
  • Discriminate the types of LC/MS according to their features and usage.
  • Compare LC/MS with LC/MSn.
  • Compare the various component options commonly found in LC/MS instruments.
    • Modes of ionization - atmospheric pressure chemical ionization (APCI) vs. atmospheric pressure electrospray (AP-ES).
    • Ion monitoring techniques - Positive vs. negative ion methods.
  • Explain the general steps involved in the operation of LC/MS systems.
  • Interpret LC/MS spectra.
  • Select operating parameters for a LC/MS separation by choosing appropriate mobile phase composition , column type, MS mode and MS parameters.
  • Compare LC/MS tuning procedures and analyse results from common tuning methods to plan LC/MS maintenance.
  • Optimise methods for qualitative and quantitative analysis using LC/MS.
  • Develop strategies for quantitative and qualitative analysis in LC/MS using common software packages.
  • Design procedures for method transfer between LC and LC/MS systems.
  • Plan and implement preventative maintenance procedures for use with LC/MS.
  • Select the correct logical steps involved in LC/MS troubleshooting.
  • Rank library generation strategies commonly used in LC/MS.
  • Illustrate the use of LC/MS in a range of applications including.
    • Determination of protein molecular weight.
    • Analysis of common macromolecules.
    • Pharmaceutical analysis.
    • Herbal analysis.
